EVI-1 expression and its impact on CAIII gene expression

This book is part of my research of MSc thesis carried out at Glasgow Caledonian University in Scotland, United Kingdom. Firstly, the book covers the aim of the project and carrying out of this project. The aim of this project was to investigate the expression of EVI-1 in Rat1neo1 and Rat15.61cells and the impact on CAIII expression. The Also is to apply RNA extraction and prepare cDNA from these to carry out quantitative Real-time PCR. Furthermore the aim is to look at the expression of EVI-1 and affected genes by extracting the protein from fibroblast cells aiming for the identification by western blotting. Introduction of plasmids to Rat1neo1 and Rat15.61 cells by transfection was another aim. Transfection with siRNA was to be aimed for. Following this, a reporter gene, luciferase, was used for testing promoter activity and its effect on CAIII expression. The goal was to inhibit EVI-1 expression in fibroblast cells by targeting the gene with siRNA. Finally was to knock down CAIII gene alone by siRNA looking at the effect of hydrogen peroxide treatment on caspase activity and apoptosis.
