For\u003c\/p\u003e\u003cp\u003ebiological macromolecules experimental phasing is one technique to\u003c\/p\u003e\u003cp\u003esolve this phase problem; it usually requires the incorporation of\u003c\/p\u003e\u003cp\u003eheavy atoms in the protein crystal.\u003c\/p\u003e\u003cp\u003eIn this thesis a new class of compounds was developed that combines\u003c\/p\u003e\u003cp\u003eheavy atoms for experimental phasing with functional groups for interaction\u003c\/p\u003e\u003cp\u003ewith biological macromolecules. Three functional groups\u003c\/p\u003e\u003cp\u003einteract with protein residues and make the compounds sticky. Three\u003c\/p\u003e\u003cp\u003ehalogen atoms provide a strong anomalous signal and may be used\u003c\/p\u003e\u003cp\u003efor experimental phasing. The halogen atoms form an equilateral\u003c\/p\u003e\u003cp\u003etriangle, which is easily recognised in the heavy-atom substructure. So far\u003c\/p\u003e\u003cp\u003eseveral novel protein structures have been solved with the iodine\u003c\/p\u003e\u003cp\u003etriangle compound.
